I see many speedtest packages in menuconfig as option.
speedtest-netperf, speedtestpp, netperf, iperf, iperf3
which one is the best one to measure download and upload and maybe some more infos?

To get best results, run speed tests from a client - not the router.
So don't need to install one of them
With the exception of speedtestapp which seems to be a client for Ookla's speedtest.net network all the others lack widely distributed public servers in the internet to test against (there are some iperf3 servers, but not that many). So for generic automated capacity testing from your router (which as @lleachii already mentioned, is not necessarily a good idea, as many routers lack the CPU to do duty as router and simultaneously sink and source capacity-test traffic) only speedtestpp seems to be suitable.
You can always install your own servers on VPS nodes under your control, but then just install the same package on both sides... (but then include iperf2 into the set of candidates, iperf2 is NOT the predecessor of iperf3 but these are two independently actively developed testing tools).
